Can keypads be wired directly to an expansion enclosure or must they be wired to the main controller?

For the OP2, yes, you can connect the keypads to the data terminals of the expansion enclosure.
 
From the 17A00 manual:
 
The Expansion Enclosure communicates with the controller using the same wires that connect the
consoles to the controller. The communications wires for the Expansion Enclosure and consoles may be
homerun to the controller or daisy-chained to each other.
